EZ tag is the optimal fusion partner for recombinant protein production. EZ tag fusion proteins can be selectively precipitated using calcium, allowing for quick and easy purification of the target protein. This minimizes the need for chromatography column work, resulting in cost and time savings
EZ tag induces formation of target protein aggregation in response to calcium ion, enabling separation from impurities. The aggregates of EZ tag-target proteins are reversibly converted into monomers simply by adding EDTA. Following cleavage of the CSQ tag by hydrolytic enzyme such as TEV proteases, only target proteins are isolated in solution by another round of calcium-responsive phase transition. Using only Ca2+, EDTA, TEV proteases and centrifugation, we purified target proteins with a high purity and high yields. Using fermentation, we can easily and cheaply obtain target proteins in grams.
By utilizing EZ tag and Z domain(Protein A) fusion proteins, antibodies can be purified without the need for Protein A column chromatography. This antibody purification technology can significantly reduce costs and time.
